Omschrijving: ASK1 Antibody: Mitogen-activated protein (MAP) kinase cascades are activated in response to various extracellular stimuli, including cytokines, growth factors and environmental stresses. A novel MAP kinase kinase kinase (MAPKKK) was recently identified and designated ASK1 (for apoptosis signal-regulating kinase 1) and MAPKKK5. ASK1 activated two different subgroups of MAPKK, MKK4 and MKK6, which in turn activated c-Jun N-terminal kinase (JNK) and p38 MAP kinase, respectively. ASK1/MAPKKK5 is activated by TNFR and Fas through the interaction with members of the TRAF family and Fas-associated protein Daxx. Overexpression of ASK1 induced apoptotic cell death, and a catalytically inactive form of ASK1 inhibited TNF-alpha -induced apoptosis. ASK1 is expressed in variety of tissues and cell lines.

APC8 is a highly conserved component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. APC/C is responsible for degrading anaphase inhibitors, mitotic cyclins, and spindle-associated proteins ensuring that events of mitosis take place in proper sequence. The individual APC/C components mRNA and protein levels are expressed at approximately the same levels in most tissues and cell lines, suggesting that they perform their functions as part of a complex. In Drosophila, silencing of APC8 results in developmental delay and pupal lethality with elevated levels of apoptosis, high mitotic index, and delayed or blocked mitosis.

IL13 has multiple effects on the differentiation and functions of monocytes and macrophages. It suppresses cytotoxic functions and induces changes in the morphology of human monocytes and in the phenotype of human monocytes and B cells by upregulating MHC class II expression. IL13 will also decrease the production of nitric oxide by activated murine macrophages, leading to impaired parasiticidal activity. Human and mouse interleukin 13 share approximately 58% amino acid sequence identity. Although human and mouse IL13 are equally active on human cells, human IL13 is much less active than mouse IL13 on mouse cells. Human IL13 and human IL4 also share approximately 30% sequence homology and have similar biological functions.

Omschrijving: IL-32 Antibody: Interleukin-32 (IL-32) was initially identified as a transcript (NK4) that is selectively expressed in lymphocytes and NK cells and whose expression is increased following activation by IL-2. It was later re-isolated from an IL-18-treated lung carcinoma cell line and re-named IL-32. IL-32 is unusual in that it does not share sequence homology with known cytokine families and is highly expressed in immune tissues, existing in at least four differentially spliced isoforms. Because treatment of human monocytic and mouse macrophage cells with IL-32 induces several proinflammatory cytokines such as TNF-alpha , IL-8 and MIP-2, and because it is also induced in human peripheral lymphocyte cells after mitogen stimulation and in epithelial cells by IFN gamma , it has been suggested that IL-32 may play a role in autoimmune and inflammatory diseases such as rheumatoid arthritis.
