Omschrijving: Chromogene media bieden een methode voor snelle identificatie van karakteristieke bacteriële enzymen. Merck biedt een groot assortiment aan vaste en vloeibare voedingsbodems die ook fluorescentiedetectie van <i>E. coli </i>mogelijk maken onder de handelsnaam Fluorocult®. Fluorocult® voedingsbodems hebben dezelfde componenten als de standaardmedia en bevatten daarnaast het substraat MUG. Sommige van de media bevatten ook tryptofaan als substraat voor een mogelijke indoolreactie om de aanwezigheid van <i>E. coli</i> verder te bevestigen. De Fluorocult® methode kan een positief resultaat opleveren binnen 24 tot 48 uur. In het geval van de chromogene media is een directe identificatie mogelijk door middel van de kleuring van karakteristieke kolonies op de voedingsbodem zelf. Een differentiatie van positieve afzonderlijke kolonies is mogelijk, zelfs bij aanwezigheid van grote aantallen bacteriën.

Incubatie: 24 ±2 uur bij 44 ±1 °C. De incubatietemperatuur heeft grote invloed op de gevoeligheid en selectiviteit van deze methode. Temperaturen hoger dan 45 °C inhiberen de groei van<i> Cronobacter</i> <i>sakazakii</i>. Temperaturen lager dan 43 °C reduceren de inhibitie van andere bacteriën.
