Several novel death receptors including DR3, DR4, DR5, and DR6 were recently identified. Cell death signal is transduced by death domain containing adapter molecules through the interaction with death domain of these death receptors. A novel TNF-R1 interacting protein was recently identified and designated SODD for silencer of death domains. SODD associates with the death domain of TNF-R1 and prevents constitutive activation of TNF-R1 signaling. TNF treatment releases SODD and permits adapter molecules such as TRADD recruiting to the active TNF-R1 complex, which activates TNF signaling pathways. SODD also interacts with DR3. SODD is ubiquitously expressed in human tissues and cell lines.

Omschrijving: APC4 Antibody: Cell cycle regulated protein ubiquitination and degradation within subcellular domains is thought to be essential for the normal progression of mitosis. APC4 is a highly conserved component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. APC/C is responsible for degrading anaphase inhibitors, mitotic cyclins, and spindle-associated proteins ensuring that events of mitosis take place in proper sequence. The individual APC/C components mRNA and protein levels are expressed at approximately the same levels in most tissues and cell lines, suggesting that they perform their functions as part of a complex. While little is known of APC4, it is thought that APC4 associates with other APC/C components APC1, APC5, and CDC23 interdependently, such that loss of any one subunit reduces binding between the remaining three.

Omschrijving: Cytokeratin-7 (CK7) is a member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. CK7 is specifically expressed in the simple epithelia lining the cavities of the internal organs and in the gland ducts and blood vessels. It is found in simple glandular epithelia, and in transitional epithelium. Epithelial cells of the lung and breast both contain CK7, but some other glandular epithelia, such as those of the colon and prostate, do not. Because the protein is found in both healthy and neoplastic cells, antibodies to CK7 can be used in immunohistochemistry to distinguish ovarian and transitional cell carcinomas from colonic and prostate cancers, respectively. Enhancement of NK cell proliferation, augmentation of non-major histocompatibility complex-restricted cytotoxicity, and induction of IFN-gamma from T cells identify ISG15 as a member of the cytokine cascade and suggest that it may be responsible for amplifying and directing some of the immunomodulatory effects of IFN-alpha or IFN-beta. ISG15 has has also been shown to function intracellularly as a ubiquitin homolog.
