De assay is gebaseerd op 5' nuclease realtime PCR-reacties om een unieke genomische sequentie te versterken in het doelmicro-organisme. PCR is een methode die wordt gebruikt om een specifieke DNA-sequentie te amplificeren, die normaal gesproken wordt geamplificeerd in een reactie met een thermisch stabiele DNA-polymerase, nucleotiden en primers complementair aan de doelsequentie. Als deze oplossing wordt verwarmd, denatureert de DNA-molecuul en wordt deze in twee strengen gesplitst. Terwijl de oplossing afkoelt, worden de primers voorverwarmd tot de doelsequenties in de gesplitste DNA-strengen en synthetiseert de DNA-polymerase een nieuwe streng door de primers uit te breiden met nucleotiden, waarbij een kopie van de DNA-sequentie wordt gecreëerd (amplicons). Als deze cyclus van denatureren, voorverwarmen en uitbreiden wordt herhaald, stijgt het aantal doelamplicons exponentieel. Bij realtime PCR worden specifieke fluorescerende sondes gebruikt om het geamplificeerde DNA te detecteren door te hybridiseren met amplicons. Deze sondes zijn aan het ene uiteinde gekoppeld aan een fluorofoor en aan het andere uiteinde aan een quencher die fluorescentie aan de andere kant onderdrukt. Als de doelsequentie aanwezig is tijdens PCR, treedt amplificatie op en degradeert de sonde, waardoor de fluorescentie stijgt. Fluorescentie wordt gemeten door een detector en de bijbehorende software maakt een grafiek van de fluorescentie-intensiteit versus het aantal cycli, waardoor de aan- of afwezigheid van het doelorganisme kan worden bepaald.

Volgens de Europese richtlijn EG 2002/86/EC moeten voedingsingrediënten worden vermeld. In het geval van vlees en vleesproducten moeten de diersoorten op het etiket worden vermeld. Daarnaast kan de authenticiteit van de diersoort uiterst relevant zijn voor consumenten, om economische, medische, culturele en religieuze redenen. Frauduleuze vervanging van duurdere diersoorten door goedkoper vlees, de aanwezigheid van vlees in producten zonder vlees (vegetarisch) en de aanwezigheid van allergenen in voedingsproducten zijn duidelijke voorbeelden die het belang aantonen van deze voorziening (Ballin, 2010; Dooley et al., 2004). Diverse methoden zijn gebaseerd op identificatie van proteïnen door middel van elektroforetische en/of immunologische methoden. Maar deze methoden zijn niet betrouwbaar voor gebruik met sterk bewerkte en verhitte producten vanwege de aantasting van de eiwitten. DNA is tijdens het verwerken stabieler dan proteïnen en ook al kan het worden gefragmenteerd door verschillende processen, staan moderne DNA-methodologieën als op PCR gebaseerde technieken nog steeds de identificatie toe van DNA van de verschillende soorten die aanwezig zijn in een monster (Lockley & Bardsley, 2000). Realtime PCR-technieken zijn speciaal ontworpen voor deze producten, omdat ook kleine fragmenten DNA nog kunnen worden geamplificeerd en geïdentificeerd met hoge gevoeligheid en specificiteit (Dooley et al., 2004).
